Description

Description: This assay employs a two-site sandwich ELISA to quantitate GNAI2 in samples. An antibody specific for GNAI2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any GNAI2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for GNAI2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of GNAI2 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Overview: GNAI2 is an alpha subunit of guanine nucleotide binding proteins (G proteins). The encoded protein contains the guanine nucleotide binding site and is involved in the hormonal regulation of adenylate cyclase. Several transcript variants encoding different isoforms have been detected for this gene, but the full-length nature of only two are known so far.
Itoh et al. (1988) demonstrated 3 distinct human G-alpha inhibitory proteins. Southern blot analysis indicated that a single copy of each gene is present in the haploid human genome.organization.Hermouet et al. (1991) demonstrated that activating and inactivating mutations of the GNAI2 gene have opposite effects on proliferation of NIH 3T3 cells
